で「その漫画は、全世界での流通販売数4億冊以上で、空前の大人気漫画である。そしてそのますます増え続けるファン層は、デビュー後20年たっても衰えを見せない。」という意味になります。

With over 400 million volumes in circulation worldwide, で「全世界での流通4億冊以上の売れ行きで」という意味になります。。
この場合の with は「～という状況で」という意味の、付帯状況を表しています。
volume は「出来高、販売数」という意味の名詞です。書籍の「巻」という意味もあります。。
in circulation は「流通している、出回っている」という意味になります。
例：quantity of money in circulation「世の中に出回っているお金の量」
動詞形 circulate は「ぐるぐる回る、流れる、広まる、流通する」です。
worldwide は副詞で「世界的に、世界中に」という意味になります。

it’s the most popular manga of all time で「それは史上最も人気のある漫画である」となります。
most ～ … of all time で「史上最～の…」という意味になります。
例：greatest film of all time「史上最高傑作の映画、空前絶後の名作」
popular は「人気のある、一般大衆にうけている」という意味ですね。

its ever-growing legion of fans is holding strong 20 years after its debut で「その増え続けるファン層はデビュー20年たった後でも衰えを見せない」となります。
ever-growing は「今でも増加している、増える一方の、ますます増加する」という意味の形容詞です。
legion は「軍団、集団」という意味で、
legion of ～ で「大勢の～、～の大群」という意味の言い回しとなります。

hold strong は「力強さを保つ、勢いを保つ」という意味の言い回しです。
例：hold a strong pace「しっかりしたペースを保つ」
