ロバートJ.ウォルディンガー氏によるTEDでのスピーチ「人生を幸せにするのは」です。

And we know that you can be lonely in a crowd and you can be lonely in a marriage, so the second big lesson that we learned is that it’s not just the number of friends you have, and it’s not whether or not you’re in a committed relationship, but it’s the quality of your close relationships that matters.

And we know that you can be lonely in a crowd and you can be lonely in a marriage, so the second big lesson that we learned is that it’s not just the number of friends you have, and it’s not whether or not you’re in a committed relationship, but it’s the quality of your close relationships that matters.
で「そしてもちろんたくさんの人に囲まれていても孤独になりえます。結婚していても孤独だということもあります。ですので、我々が学んだ第二の大きな教訓というのは、単に友達の数ではなく、また決まった人がいるかどうかでもなく、あなたが置かれたその新密な関係の質が、大切だということです。」という意味になります。

And we know that ～ で「私たちは～と言うことを知っている」です。
周知の事実だということです。

(that) you can be lonely in a crowd で「人は孤独になり得る、群衆の中で」です。
この場合の you は総称人称の you で「人は、人間は～、誰でも～」といった意味です。
lonely in a crowd で「人混みの中で孤独な」です。
つまり、周りに人がいっぱいいても孤独を感じることがある、ということですね。

and you can be lonely in a marriage で「また、人は結婚生活の中でも、孤独になり得る」です。
in a marriage で「結婚生活で、婚姻関係の中で」です。

so「ということで～」
the second big lesson that we learned が主語で「我々が学んだ第二の大きな教訓」です。
is that ～「は～ということだ」

どういうことかというと、
(that) it’s not just the number of friends you have, で「（重要なことは）単にあなたが持つ友達の数ではない」です。
この it は、強調構文の It ～ that … 文型の主語となります。
間が長いのでわかりづらくなっていますが、
It is ～ that matters.「重要なことは～だ」と最後の that matters に繋がっています。

そして、not just ～ but …「～だけではなく…だ」という文型も入っています。

単に友達の数だけじゃなく、さらに、
and it’s not whether or not you’re in a committed relationship,「また、パートナーの有無でもない」です。
whether or not ～ で「～であるかどうか」という意味の言い回しです。
(whether or not) you’re in a committed relationship, で「信頼しあった関係にある（かどうか）」です。
committed relationship は直訳だと「献身的な関係」ですが、
慣用的に「婚姻関係、恋愛関係、パートナー関係」という意味合いです。
お互いに commit「全てを捧げる」ような関係ですね。
例： committed to ～「～に専心する、～と固く結ばれている」
前半に出た in a marriage の言い換え表現になっています。

but ～
ただ単に友達の数でもなく、結婚しているかどうかでもなく、
it’s the quality of your close relationships で「（大事なのは）その親密な関係の質である」です。

that matters. は it に繋がって「大切なのは～だ」です。
matter は動詞で「大事である、大切である、どうでもいいことではない」という意味です。
例：Black Lives Matter「黒人の命は軽くない／命に白人も黒人もない」

それではまず文節に分けて日本語に訳し内容を理解して下さい。

And we know that
そして周知のとおり

you can be lonely in a crowd
群衆の中にあっても人は孤独になり得ます

and you can be lonely in a marriage,
また結婚していても孤独になり得ます

so the second big lesson that we learned
ということで、我々が学んだ2番目の大きな教訓は

is that
～ということです

it’s not just the number of friends you have,
（大事なのは）単に友達の数ではなく

and it’s not
また～でもない

whether or not you’re in a committed relationship,
生涯決まった相手がいるかどうか

but
ではなく～だ

it’s the quality of your close relationships
（大事なのは）質である、その親密な関係の

that matters.
大事なのは

今度は「頭の中で」一行ずつ日本語に訳しながら…

And we know that
