で「しかし手段は変われど、我々の目的は変わりません。その目的とはつまり、一人一人すべてのアメリカ人の努力と決断力に報いる国家であることです。それこそがこの時代が求めているものです。それこそが我々の信条に本当の意味を与えるものです。」という意味になります。

But「しかし」
while the means will change, で「手段は変わるだろうけれども、」です。
while ～ は「～である一方で、～ではあるが」という意味の接続詞。
our purpose endures で「我々の目的は不変である」です。
endure エンデュアーは「持ちこたえる、変わらないままでいる」という意味の動詞です。
change に対応した意味合いですね。

コロン（:）で、「それはつまり～」という意味です。
our purpose のことで「我々の目的とはつまり～」

a nation that ～ で「～をする国家」です。
我々が目指すものは「～をする国家」ということです。
何をする国家かというと、
(that) rewards the effort and determination of ～ で「～の努力や決断力に報いる（国家）」です。
reward リウォードゥで「～に褒美を出す、～に報いる」という意味の動詞です。
(the effort and determination of) every single American. で「一人一人すべてのアメリカ人（の努力や決断力）」

That is what this moment requires. で「それこそがこの時が求めているものだ」です。
that is what ～ で「それこそが～するものだ」です。
this moment は「今まさにこの時、現時点」です。

同じ文型で、
That is what will give real meaning to our creed. で「それこそが、私達の信条に本当の意味を与えるものだ」です。
give ～ to … で「～を…に与える」です。

それではまず文節に分けて日本語に訳し内容を理解して下さい。
