で「どの世代にも、その時代を象徴する偉業があります。30万人以上の人々が、ひとりの人間を月に送るというプロジェクトに関わりました ―― その中にはあの清掃員も含まれていました。何百万人ものボランティアが、世界中の子どもたちにポリオワクチンを接種しました。さらに何百万人もの人々が、フーバーダムやその他の偉大な建造物を造り上げたのです。」という意味になります。

Every generation has its defining works. で「すべての世代は、その世代を象徴する偉業を持っている。」です。
defining はここでは「象徴的な、それを決定づけるような」という意味の形容詞。
動詞形 define は「～を定義する」です。

More than 300,000 people「30万人を超える人々が」
worked to put a man on the moon「一人の人間を月に送るために働いた。」
– including that janitor.「あの清掃員の彼も含めて」です。
スピーチの前半で出てきた janitor のことを指しています。

Millions of volunteers immunized children で「数百万人ものボランティアたちが、子供たちに予防接種をした」
immunize [イ]ミュナイズで「～に免疫をつける、予防接種する」です。
形容詞形 immune イミューンで「免疫の」
例：immune system「（体の）免疫系、免疫システム」
(children) around the world「世界中の（子供たちに）」
against polio. で「ポリオに対抗して（予防接種をした）」

Millions of more people で「さらに数百万の人々が」
built the Hoover dam で「フーバーダムを建設した」
and other great projects. で「また、その他の偉大な建造物を」です。
Hoover dam はアメリカで最大級のダムです。
1930年代の世界恐慌時に雇用創出のために始まった公共事業で、フーヴァー大統領にちなんで名づけられました。
