The Ise Jingu shrine で「伊勢神宮」のことです。
shrine シュラインは「神社、神殿、聖廟」といった意味の名詞です。
意味的には「聖なるものが宿る場所、建物」というイメージになります。

geographical and spiritual heart of Ise で「地理的にも精神的にも伊勢の中心地」です。
heart of ～ で「～の中心、～の心臓部、～の核心」という意味になります。
geographical は「地理的な」という意味の形容詞。
名詞形は Geography ジ[ア]グラフィ「地理」です。

making up around a fifth of the town’s total area で「街の全エリアの1/5を占めていて」です。
make up は「～を構成する、形作る、～を占める」という意味の言い回しです。
例：make up a large share of ～「～の大部分を占める」
ちなみに make up には「化粧する」や「話をでっちあげる、ねつ造する」という意味もあります。
化粧するとねつ造するが同じ表現というのは実に興味深いですね。

around は「約～」という意味です。about とも言い換えられます。

a fifth of ～ で「～の5分の1」という意味になります。
分数や倍数の言い方は、とてもよく使われますし、覚えておくと会話でも便利です。
分数：a third of ～（one-third of ～）「～の3分の1」、three-fifths of ～「～の5分の3」
倍数：two times bigger than ～「～より2倍大きい」

それではまず文節に分けて日本語に訳し内容を理解して下さい。
