で「江戸城の外堀に位置していたため、神楽坂は大きな歓楽街として常に人気を博してきた。そしてかつては芸者が多くいる場所だった。」という意味になります。

Due to ～ は「～に起因して、～のために」という意味の言い回しです。
起因・原因を意味します。

its location on the outer moat of Edo Castle で「江戸城の外堀に位置すること」です。
location は「場所、位置、所在地」です。
moat はお城の周りなどに掘られる「掘り、お堀」という意味です。
outer moat なので「外堀」ですね。

enjoyed popularity as a major entertainment district で「大きな歓楽街として人気を博する」です。
enjoy popularity で「人気を博す、好評である」という意味の慣用句です。
人気がある状況を楽しむ→人気がある状況を謳歌するという意味合いですね。
例：She enjoyed her popularity at the cost of her privacy.「彼女はプライバシーを犠牲にして人気者になった。」
as a major entertainment district で「大規模な歓楽街として」です。
entertainment district で「歓楽街」という意味になります。
いわゆる繁華街・大人の遊び場的な街のことを意味します。

was formerly home to hundreds of geisha で「かつては芸者の生息地であった」です。
formerly は「かつては、以前は」という意味の副詞です。
形容詞形は former で「かつての、前～」です。
例：former mayor「前市長」
be home to ～ で「～の本拠地、～の生息地、～が住む場所、～のメッカ」という意味です。
例：home to jazz「ジャズのメッカ」、home to Japanese culture「日本文化の古里」

それではまず文節に分けて日本語に訳し内容を理解して下さい。
