で「最終的に人を評価する基準は、その人が、快適で楽なときではなく、課題と論争の中でどういう立場をとるかである。」という意味になります。

The ultimate measure of a man で「人を見抜く究極の方法」という意味です。
ultimate は「究極的な、最終的な」という意味の形容詞です。
例：ultimate aim「究極的な目標」
measure メジャーは「寸法、計量器、基準原器」という意味です。
動詞では「測る、評価する、測定する」という意味になります。
大きさ・重さともに使います。

not ～ but …「～ではなく…である」の形です。

not where he stands in moments of comfort and convenience, で「快適で楽なときにどんな立場をとるかではなく」です。
where he stands で「どこに立つか、どういう立場をとるか、どんなスタンスでいるか」です。
in moments of comfort and convenience で「快適と便利さの時に」です。
in moments of ～ で「～な時に」という意味の言い回しです。
例：in moments of pain「苦しい時に」

but where he stands at times of challenge and controversy. で「（ではなく）課題と論争の中でどういう立場をとるかである」です。
at times of ～ で「～な時に」です。
in moments of ～ の言いかえですね。
challenge and controversy で「課題と問題」です。
challenge は「挑戦」という意味もありますが、今回は「課題、問題」という意味です。
controversy は「議論、論争」という意味です。
例：controversial point「問題点、賛否両論のポイント」

それではまず文節に分けて日本語に訳し内容を理解して下さい。

その人がどういう立場をとるか（ではない）
