で「今日、バスルームの床に黒くて大きい蜘蛛がいるのを見つけた。私は即座に便座に飛び乗って、滑り落ちて、壁に頭をぶつけた。この話の何が面白いかって？それは私の髪の毛の束だった。F*ck my life!」という意味になります。

I saw a big, black spider on the floor of my bathroom. で「私は見た、大きくて黒い蜘蛛を、バスルームの床に」です。
a big, black spider で「黒い大きな蜘蛛」です。
英語での形容詞の並び順はしっかり覚えておきましょう。

I promptly jumped up on the toilet seat で「私は即座に飛び乗った、便座に」です。
promptly は「即座に、すぐに、素早く」という意味の副詞です。
例： promptly respond to ～「～に即答する」
toilet seat で「便座」ですね。
seat スィートは「椅子、腰掛、尻のを乗せる面」という意味になります。
発音は seat スィートゥとなります。
sheet シートではないので注意しましょう。

slipped off, and slammed my head against the wall. で「滑り落ちて、頭をガツンと壁にぶつけた」です。
便座に急いで飛び乗ったので、足がすべって落ちてしまったということですね。
slam スラムはドアなどを強く閉めるときの音、バタンという音からできた動詞で「ドカンと強く打つ」という意味になります。
例：slammed the phone down「受話器をガチャンと切る」、slam a car door「車のドアをバタンと閉める」

The best part? It was my tuft of hair. で「何が面白いかって？それは私の髪の毛の束だった」です。
黒い蜘蛛だと思ったものは、髪の毛の束だったということです。
The best part? で「この話の何が面白いかって？」という意味の言い回しです。
best part は「物語のいいところ、佳境、見どころ」という意味合いです。
tuft タフトゥは「束、房、茂み」という意味です。
例：tuft of grass「草むら」
