で「ステージ・メイクや役柄などの内側にいるのは、君だ。そして、思うに観客が本当に興味があるのも、君だ。君がどう対処するか、作家が君の前に用意した、その状況、その障害、その困難に。」という意味になります。

Inside of all the makeup and the character and makeup, it’s you, で「メイクアップ、役柄やメイクアップのの内側に」です。
makeup はここでは俳優としての舞台化粧・ステージメイク・かつらやパッドなどの事でしょう。
character は「登場人物、キャラクター、役柄」です。
その内側（にいるのは）
it’s you,「それはあなただ」です。
and I think ～「そして～だと思う」
that’s what the audience is really interested in. で「それこそが観客たちが本当に興味があるものだ（と）」です。
audience は集合的に「観客、観衆、聴衆」です。

.. you, で「（つまりそれは）あなた」です。
how you’re going to cope with the situation, で「あなたがシチュエーションにどう対処するか」
the obstacles, the troubles で「その障害に、その困難に」
(that) the writer put in front of you. で「作家があなたの前に用意した（そのシチュエーション、障害、困難）」

それではまず文節に分けて日本語に訳し内容を理解して下さい。
