で「私は、完璧な結末を望んでいました。でも今は、つらい経験を通して学んだのです。韻を踏まない詩もあれば、はっきりした始まり・途中・終わりを持たない物語もあるのだと。人生とは、先のことがわからないまま、変わらざるを得ず、その瞬間を受け止め、できるかぎり最善にしていくことなのです。そんな“あいまいさ”もまた、味わい深いものなのです。」という意味になります。

I wanted a perfect ending. で「私は完璧な結末を望んでいた。」です。
過去形を使うことで「でも今はそうではない」という含みを持たせられます。

Now I’ve learned, the hard way, that ～ で「でも今は、私は～だということを、つらい経験を通して学んだ」です。
learn the hard way that ～ で「つらい経験を通して～だということを学ぶ」という意味の言い回しです。
(that) some poems don’t rhyme, で「韻を踏まない詩もある（ことを）」
, and ～「また～」
some stories don’t have a clear beginning, middle, and end. で「明確な始まり・真ん中・終わりがない物語もある（ことを）」

Life is about ～ で「人生とは～だ」です。
この about ～ は「～について」ではなくて、「～につきる、～が目的だ」という意味です。
例：Science is about knowing.「科学の目的は知ることである。」
not knowing,「知らないこと」
, having to change,「変わらなければならないこと」
, taking the moment and making the best of it,「その瞬間を受け入れて、それを最大限に活かすこと」です。
make the best of ～ で「～を最大限活かす」という意味のコロケーション。
, without knowing ～「～を知らない状態で」
what’s going to happen next.「次に何が起きるかを」

Delicious ambiguity.「味わい深い曖昧さ」です。
ambiguity アンビ[ギュ]イティは「曖昧さ」という意味の名詞。
形容詞形は ambiguous アン[ビ]ギュアス「曖昧な」です。

それではまず文節に分けて日本語に訳し内容を理解して下さい。
