で「もちろん、このすべてが効力を発揮するところは、とりわけ何かとても大事なことをする時です。それはこんな風にステージに立ってスピーチをする時かもしれません。プロポーズしたり、賃上げ交渉、または結婚式でのスピーチの時かもしれません。」という意味になります。

where this all comes into play most of all is ～ で「このすべてがとりわけ効力を発揮するところは」となります。
this comes into play で「これが効果を出し始める、これが作用し始める」です。
come into play で「効果が出始める」となります。
come into は「～になり始める」という意味の言い回しです。
play は「再生される、音などが出る、上映される」です。
most of all は「とりわけ、なによりも」と言う意味の副詞句です。

前回まで話してきた、
register「声域」
timbre「声色・声質」
prosody「韻律」
pace「ペース」
pitch「声高」
volume「声量・音量」
の「効果が現れる時」ということですね。

is when you’ve got something really important to do で「あなたが何かとても重要なやることがある時だ」となります。
you’ve got ～ で「～を持っている」と言う意味になります。
something really important to do で「とても重要なやるべきこと」です。

「重要なやるべきこと」とは何かというと。

It might be ～ で「それは～かもしれない」
standing on a stage like this and giving a talk to people. で「こんな風にステージに立つこと、そして人にお話をすること」です。
give a talk は「講演をする、スピーチする」と言う意味の言い回しです。
give a speech とも言います。

It might be proposing marriage, で「結婚の申し込みかもしれない」です。
propose marriage で「結婚の申し込みをする、プロポーズする」です。

asking for a raise で「給与を上げるよう頼むこと」です。
raise レイズは「上げること、賃上げ」です。
例：get a raise「昇給する」
ask for ～ で「～を要求する」と言う意味の句動詞です。
ask は「尋ねる」と言う意味もありますが、この場合は「要求する、頼む」と言う意味になります。

それではまず文節に分けて日本語に訳し内容を理解して下さい。
