I have a dream that my four little children will one day live in a nation where they will not be judged by the color of their skin but by the content of their character.
で「私には夢がある。いつの日か、わたしの４人の子供たちが、肌の色で判断されず、彼らの人格という中身によって判断される国で人生を送るという夢が。」という意味になります。

I have a dream that ～ で「私には夢がある、～という夢が」です。
「～という」という意味の同格のthat になります。
どんな夢かというと、
my four little children will ～「わたしの４人の幼い子供たちが～」
one day「いつの日か」
live in a nation (where) ～ で「～な国家に暮らす（という夢が）」です。
どんな国家かというと、関係副詞 where で説明が入ります。
(where) they will not be judged by the color of their skin で「（その国家では）彼らは肌の色で判断されることの無い」です。
but ～「そうではなくて～」
by the content of their character. で「彼らの人格という中身によって（判断される）」です。
not ～ but … の形で「～ではなくて…」という意味の言い回しです。
(by) the content of their character. は「中身（によって）、彼らの人格という（中身）」です。
character には「キャラクター、性格、気質」という意味外にも、「品性、人格、人柄」という意味もあります。
例：have character「品位がある、人格者である」

それではまず文節に分けて日本語に訳し内容を理解して下さい。

I have a dream that
私には（～という）夢がある

my four little children will
私の4人の幼い子供たちが

one day
いつの日か

live in a nation
～な国家で暮らす（という）

where
（その国家では）

they will not be judged
彼らは～で判断されない

by the color of their skin
肌の色で

but
ではなくて

by the content of their character.
人格という中身で（判断される）

今度は「頭の中で」一行ずつ日本語に訳しながら…
