で「地元住民たちは長い間、高速道路がその49メートルの橋（日本橋）の景観を損ねていると抗議していた。かつての国交大臣は2001年に、高級ショッピング地区である日本橋エリアの高架道路を別ルートにすると誓約したが、その計画は巨額のコストのために2018年まで進展しなかった。」という意味になります。

Local residents で「地元住民、地域住民」という意味になります。
local は「その地域の、地元の」という意味の形容詞。

had long protested で「長く抗議してきた」です。
過去完了形になって、ある時点まで～してきた、という意味になっています。
protest that ～ で「～だと抗議する、～だと断固主張する」という意味の言い回しです。
protest は「～に抗議する」という意味もありますが、ここでは「～だと断固主張する」という意味で使われています。
例：protest one’s innocence「自分の無罪を主張する」
何と断固主張していたかというと、
(that) the expressway spoils the view of the 49-meter bridge. で「その高速道路が～を駄目にしていると、その49メートルの橋の景観を」です。
the expressway で「その高速道路」です。日本橋の上に走る首都高のことですね。
spoil ～ は「～を甘やかす」という意味もありますがここでは「ダメにする、台無しにする」という意味です。
the 49-meter bridge は Nihonbashi の言い換え表現（パラフレーズ）です。

A former land minister in 2001 で「2001年の元国交大臣」です。
land minister で「国土交通省大臣」です。国交大臣の正式名称は、Minister of Land, Infrastructure, Transport and Tourism となります。
pledged to reroute the overpass で「～すると誓った、その高架道路を別ルートに直すと」です。
overpass は「高架、高架道路」という意味の名詞です。

in the Nihombashi area, an upscale shopping district, で「日本橋エリア内で、高級ショッピング地区（である日本橋エリア）」です。
upscale は「高級な、上流階級の」という意味の形容詞です。
例：upscale condo「高級マンション」、upscale hotel「高級ホテル」

but ～「しかし」
the plan did not proceed で「その計画は進展しなかった」です。
the plan とは高架を別ルートに直す計画のこと。
proceed で「進む、進展する、前進する、はかどる」です。

due to the huge cost. で「膨大なコストのせいで」です。
due to ～ で「～のせいで、～のために」です。
例：due to a family crisis「家庭の事情で」
