で「しかしその100年後、黒人たちは今だ自由ではない。100年後、黒人たちの生活は今だ悲しいことに、人種隔離という手枷と差別という鎖によって不自由を強いられている。」という意味になります。

the Negro still is not free. で「黒人は今だ自由ではない」です。

One hundred years later,「それら100年後」
the life of the Negro is still ～ で「黒人の生活は今だに～だ」
sadly「悲しいことに」
crippled by ～ で「～によって手足を不自由にされている」です。
cripple は「～に手足を不自由にする、不具にする、活動不能にする」という意味の動詞です。
名詞では「（手足の）身障者、廃人」などという意味になります。
例：sexual cripple「性的不能者」、crippled economy「駄目になった経済」
by the manacles of segregation で「人種隔離という手かせによって」
manacle は「手かせ、手錠」です。
segregation は「隔離」という意味から「人種差別による隔離」という意味でも使われます。
白人と黒人が同じ学校にいったり、同じレストランで食事をすることが許されなかったリしたことを指します。
動詞形は　segregate で「～を分別する、～を分離する」です。
例：segregate waste for reuse「廃棄物を分別して再利用する」
and「また～」
(by) the chains of discrimination. で「差別という鎖によって」です。
