で「そして、驚くことなかれ、25万の人々がちょうどその日、ぴったりの時間に彼の話を聞きに姿を現しました。彼のために集まった人がどれくらいいたか？ゼロです。彼らは自分自身のためにやってきたのです。」という意味になります。

lo and behold は「何たることか！」という意味合いの掛け声です。
lo は間投詞で「見よ！」という意味合があります。

250,000 people showed up で「25万の人々が姿を現した」です。
show up は「（会場など）姿を現す、やってくる、来場する」です。
on the right day で「ぴったりのその日に」です。
at the right time で「ぴったりの時間に」となります。
この場合の right 「ちょうどの、ぴったりの」という意味の形容詞になります。
例：right word「ぴったりの言葉、ちょうどいい言葉」

to hear him speak で「彼が話すのを聞くために」です。
hear＋人＋動詞原型で「人が～するのを聞く」となります。

How many of them showed up for him? Zero. で「彼らのうち何人が彼のためにやってきたか？ゼロです。」です。
